About *Sorghum Ji2055 *

Ji2055 is an inbred line of Sorghum bicolor originating in China, with a genome size of 722.96 Mb. Known for its red seeds, Ji2055 has contributed to the release of numerous commercial sorghum varieties and is recognized for its agronomic traits, including disease resistance and environmental adaptability (Yuan et al., 2024). This genome serves as an essential resource for studying sorghum’s genetic diversity and potential for improvement.

Assembly

  • DNA Extraction & Sequencing: High molecular weight (HMW) DNA was extracted and sequenced using PacBio HiFi, Oxford Nanopore (ONT) ultra-long reads, Illumina short reads, and Hi-C data. The contigs were assembled using Hifiasm, and Hi-C data was used for scaffolding.
  • Genome Assembly: The T2T assembly produced a 722.96 Mb genome, with all gaps closed and assembly errors corrected. The assembly is highly contiguous and gap-free, covering centromeres and telomeres with high accuracy.

Annotation

Repeats & Gene Models: The genome contains 65.22% repetitive elements, primarily retroelements and DNA transposons. A total of 36,950 protein-coding genes were annotated, with the majority of gene models supported by RNA-seq data.Compared to BTx623, the Ji2055 genome captures distinct sequence variations, especially in centromeric regions, offering insights into sorghum's genetic diversity. Ji2055-T2T improves the understanding of sequence composition and gene function in complex, repetitive regions.
